Dehydrated culture media is one of the greatest developments in the field of Microbiology and all the credits go to Mt Robert Koch, who actually used a culture media that is solid on which bacteria would grow.

What is Culture Media?

All the micro organisms or microbes require nutrients in order to enhance their growth. These nutrients can be supplied to the microbes through solid culture media or liquid culture media. Agar is the nutrient that is used in solid standard media. When the microbiologists would like to inhibit the growth of microorganisms by simultaneously encouraging the growth of other microbes, then a special type of media is used which is termed as selective media. The selective media has special elements that help in the growth of some kind of microbes and not all of them.

Advantage of Dehydrated Culture Media:

During the preparation of complex culture media, it is always better to use readymade standardized dehydrated media in order to ensure good performance and high rate of reproducibility. Most of the time, this option is less expensive compared to buying chemical constituents individually. Also, some chemicals are hard to find in India and some chemicals will not be sold in low quantities. They should be bought in bulk and they will usually have short shelf life.

Dehydrated culture media is hygroscopic. It has the ability to absorb water. The manufacturers manufacture this culture media in a way it absorbs water. Therefore, the moment it is exposed to moisture, the dehydrated culture media becomes unfit for use. The microbiological and chemical properties get changed as a hard mass is formed when exposed to moisture. Therefore when dehydrated culture media is being used, good precautions should be considered to prevent deterioration of the material. In tropical countries where the content of moisture is high in atmosphere, this is a cause of concern.

Here are some precautions you can follow to enable proper use of dehydrated culture media.

1. Weigh the medium very quickly and cap the bottle tightly as soon as possible. If some quantity of it is left over after your use, do not add it to the bottle.

2. Seal the cap of the bottle with an adhesive tape. If you cannot seat the bottle for some reason, place the bottle in an air tight container after squeezing out the air in the container by whatever means.
