No at all,this has nothing to do with ranking. A good BACK LINK will give you rank. There are other factors that makes a website get ranking. Make sure your domain will expire in next 5 years,this will make Google think you serious and allow people able to comment on your website. Comment is very important

If that's the main site, then put it in the root of the domain. Yes, it will affect, but if You set the redirect as permament redirect, Google will index the new Urls. Secont - better option is to install Your site to root directory.
