Pharmaceutical Pellets manufacturing process in detail

by Sanjeev Yadav SEO Consultant
Pellets are small, spherical particles made by the accumulation of fine powders or granules of drug substances. This process is done using appropriate processing equipment. Pharmaceutical pellets come with many additional benefits over conventional tablets.

Reason for wide acceptance
The reason for their wide acceptance lies in their versatility that it can be used to target tissue in order to maximize the drug delivery at the desired site of action within the given period of time in the gastrointestinal tract.

Pelletization Techniques
As compared to single dosage form pellets are technically more complicated to manufacture, but it possesses a number of advantages also.

Pelletization has become quite famous due to its various advantages over other similar techniques like the uniformity of dose and flexibility in dosage form design. There are a number of Pelletization techniques currently used by numerous pharmaceutical pellets supplier in Gujarat.
• Extrusion spheronization
• Layering Technique
• Cryopelletization
• Hot Melt Extrusion
• Freeze Pelletization

Pelletizing Process Principles
•    Coating of Pellets
The coating is the process of applying coating material by spraying onto the moving bed of pellets with concurrent use heated air which facilitates evaporation of the solvent. There are types of coating available.
•    The liquid layering of pellets:
The multi-functional coater and top spray coater systems can all be used to make pellets by layering the active material onto an inert core. Non-pareil starter pellets are sprayed with a solution or suspension of the active material and dried simultaneously.
•    Powder layering of pellets:
When the present composition is in powder form, Pelletization can be attained by spraying starter pellets with the active powder along with a liquid binder solution at the same time. The layered pellets are then dried.

The main objectives of the coating of pellets are as follows:
1. In order to mask the taste, odor, and color of the drug.
2. It avoids inactivation of drugs in the stomach.
3. It prolongs the dosing interval.
4. It improves the patient’s compliance.
5. It improves the appearance and texture of the dosage form.

Melt granulation pelletization:
Melt pelletization can be done using the Pellet Processor or the Single Pot Processor. The active and binder powders are mixed and heated to a temperature above the melting point of the binder.

This is a very fast, truly one-pot process used by wholesale pharmaceutical pellets supplier in India during which a heated jacket or optional microwave energy can be used to melt the powders. This helps to form pellets from 0.5–2.0 mm in diameter.

Wet granulation pelletization:
The active substance is mixed with microcrystalline cellulose (5–30%) and the mixture is granulated with water or an organic solvent. During the process, the granules are compacted and spheronized.

Extremely round pellets can be produced at batch capacities up to 150 kg.

Extrusion and spheronization:
This is a turnkey pellet production plant that combines a mixer/granulator, extruder, and spheronizer in a single, integrated process. Wet powders constantly pass through a low-shear radial extruder. Then the extrusions are split and formed into pellets in the spheronizer.
