Cases in which Intra Uterine Insemination is ineffective:
Blocked or damaged uterine tubes.

- Ovarian failure.
- Severe seminal fluid anomaly.
- Severe endometriosis.

There are basic investigations that should be done for the woman before the procedure is performed and it includes ovulation control and the patency or franchise of the fallopian tubes.

IUI Sample preparation: Preparation for Intra-Uterine Insemination 
IUI Sample preparation include-

1 Controlling Ovulation: 

Previously, ovulation was monitored by recording body temperature and noting any change in vaginal discharge, or by doing blood and urine tests on Days 12-16 of the cycle. Now the ovulation check is done by vaginal ultrasound after the woman is given ovarian stimulant medication to ensure the maturity of the follicles (the eggs) and the doctor usually prefers to have at least 1 -3 adult follicles to give the HCG injection. The seed preparation usually takes 1-3 hours and IUI is done in the time that is decided by the doctor. 
 
2-Preparation of the seed sample:

A seed sample can be produced by masturbation at home, or in a special room in the hospital, after 2-5 days of abstinence. The semen is then washed in the laboratory, thus isolating normal motile sperm from abnormal immotile sperm. The sperm is then separated from other components of the semen and concentrated in small quantities. Different media and techniques can be used for the Preparation of the seed sample. Sperm Preparation takes around 30-60 minutes.
